There is something wrong when 5 year olds know the word "hooker." There's a parenting fail in there somewhere. Either you exposed her to it (she overheard you/dad/your friend, she heard it on an inappropriate TV show, etc) or she heard it from her classmates in kindergarten who have lazy parents.
This sounds fake to me. A 5 year old wouldn't know what a prostitute is, let along know the slang 'hooker' for it.

A good point, but I would disagree with you on that, you'd be surprised the things children know at absurdly young ages. She could have heard the word any number of places (School, TV, Radio, her parents, etc). Granted she probably doesn't know what it means, like most small children, but not knowing what something is doesn't stop children from talking about it.
HA! I knew it! Barbie really is the root of all evil! Don't worry too much, she probably heard the word at school or on the radio or something, chances are she has no idea what a hooker actually is and she's just saying it for the hell of it. I assume afterwords you sat down and talked to her about why she shouldn't say that, so there shouldn't be anything to worry about.
