Well now you have the practice to make another one in no time :D
I've seen framed puzzles with a single piece missing for effect. If people have done it on purpose; why couldn't you have?
Put your name or "I love you grandma" or something in the place where the missing piece is and make it seem like a heartfelt idea rather than an accident.
Or just "dog ate it :(". Makes for a pretty funny story.
I highly doubt any store would accept that return. They would have you call the manufacturer.

this happened to my mom one time. she put together a 1000 piece puzzle and the last piece went missing. she went and bought another of the same puzzle and went through the whole damn thing to find that one piece.

What my mom does when we lose pieces is she cuts out some poster board in the same shape as the missing piece, and then draws a new one to match the rest. I don't know how your art skills are, but it's probably worth a shot
Try pasting the puzzle on cardboard and paint the last piece in. That is what we did once.
