Punish the dog then and there while they're doing the unwanted act, and they'll understand. Also, get another puzzle, put the dog up or outside for as long as it takes to finish so there's 0 chance it can screw up your work again. Feed, water, and potty it before locking it up. inb4 "wahh abuse!"
Depending on the location and your grandmother's sense of humor, I like the idea of a tiny handwritten note in the blank spot, even just a simple "oops." Otherwise, definitely try contacting the manufacturer.
Buy another puzzle at Goodwill or the Salvation Army

What my mom does when we lose pieces is she cuts out some poster board in the same shape as the missing piece, and then draws a new one to match the rest. I don't know how your art skills are, but it's probably worth a shot
Try pasting the puzzle on cardboard and paint the last piece in. That is what we did once.
