If I had to guess, I would guess the rooms have an electronic lock on the door with a card type key. That is most likely linked into a system the hotel that activates the card key, etc. I am just guessing that either your door lock is acting up or the hotel security system is AFU. (A) Usually there is a mechanical safety lock on the door inside the room. Nobody can get through that if it’s engaged, regardless of what the electronic lock is doing. (B) Call the front desk, they should answer no matter what time it is day or night. Explain the problem you have. They will probably offer to move you to a different room. They are obliged to provide a safe and secure room. (C) Whatever you do, do not watch or read “1408” - A Stephen King story about an evil hotel room.

Put something there to block it. Your hotel dosen't have a lock though? I'd get a refund and go elsewhere for safety reasons if possible.
